ScreenCell® Isolation Devices

ScreenCell® MB kit is devoted to molecular biology studies. Each kit contains a set of single use DNAse- and RNAse-free filtration devices individually packed, together with a specific buffer; a collection tube is also included. ScreenCell® MB kits are available for isolating circulating rare cells alive through filtration devices.

Before filtration, 6 ml blood samples must be diluted in 1 ml of the ScreenCell® LC dilution buffer, followed 2 minutes later  by addition of 1.6 ml of adequate tissue culture medium, allowing isolation of live cells.

At the end of filtration, the nozzle/holder of the ScreenCell® MB is unclipped and removed from the filtration module. The capsule-filter is then ejected and either inserted inside the upper inner part of a nuclease-free Eppendorf® tube, by pushing down a rod located at the bottom part of the filtration device. The capsule-filter may also be ejected into a well of a 24 well tissue culture plate before insertion inside an Eppendorf® tube.

Then the ScreenCell MB filtration unit allows either DNA or RNA extraction directly from cells isolated on the filter before or after cell culture (the cupule-filter containing the cultured cells is then transferred with a nuclease-free forceps into an nuclease-free Eppendorf® tube.

An adequate volume of lysis buffer is added into the capsule-filter which is then closed with the Eppendorf® tube cap in order to extract DNA or RNA directly from cells isolated on the filter. Following incubation at the appropriate temperature, the Eppendorf® tube containing the capsule-filter is centrifuged and the capsule-filter removed and discarded. The flow-through is either stored in the closed Eppendorf® tube or use immediately, to conduct further molecular biology procedures.

ScreenCell® CC kit is devoted to cell culture. It contains a set of single use sterile filtration devices individually packed, together with a specific buffer and a filtration tube. ScreenCell® CC isolation device contains a circular filter, located inside the bottom part of the filtration module and capped by a removable nozzle/holder.

Before filtration, 6 ml blood samples must be diluted in 1 ml of the ScreenCell® LC dilution buffer, followed 2 minutes later  by addition of 1.6 ml of adequate tissue culture medium, allowing isolation of live cells.

At the end of filtration, the nozzle/holder of the ScreenCell® CC device is unclipped and removed from the filtration module. The circular filter is then released into a well of a 24 well tissue culture plate, by pushing down a rod located at the bottom part of the filtration device.

Adequate tissue culture medium and growth factors are added into the well. The multiwell plate is then closed and incubated under defined conditions. Cytological studies can then be conducted directly on the filter.

ScreenCell® CY kit is devoted to cellular biology studies. It comprises a set of  single use filtration device individually packed, for isolation of circulating rare cells together with a specific buffer; a collection tube is also included.

Before filtration, 3 ml blood samples must be diluted in 4 ml of the ScreenCell® FC filtration buffer for fixed cells. An additional 1.6 ml of 1X PBS is filtered for removing debris from the filter.

At the end of filtration, the nozzle/holder of the ScreenCell® CY device is unclipped and removed from the filtration module. The filter is then released onto a standard microscopy glass slide by pushing down a rod located at the bottom part of the filtration device. Cytological studies such as staining, cell enumeration, immunocytochemistry, FISH assays, etc… can be conducted directly on the filter, the filtration area being delimited by a more rigid O ring.

 ScreenCell® Dilution Buffers

ScreenCell® Fixed Cells (FC) Dilution Buffer

The ScreenCell® FC dilution buffer is essentially for pathocytological studies, including cell enumeration, cytology, immunochemistry and FISH assays (accessorily, molecular biology applications are possible through laser microdissection). It allows lysis of RBCs before filtration while preserving other cells. This buffer is for cytology, immunochemistry and molecular biology applications on fixed cells.

ScreenCell® Live Cells (LC) Dilution Buffer

The ScreenCell® CC dilution buffer is for isolation of live cells for cell culture directly on the filter after filtration through: (i) the ScreenCell® CC filtration device for further cytological applications; (ii) the ScreenCell® MB filtration device for further molecular biology (DNA/RNA extraction) applications. It allows a mild lysis of red blood cells (RBCs) before filtration while leaving other cells alive.

ScreenCell offers hands-on instrument training sessions as well as applications-based courses on the use of the ScreenCell® devices and protocols in cell biology, cell culture and molecular biology which apply to rare circulating cells.
